Therapeutic angiogenesis promotes efficacy of human umbilical cord matrix stem cell transplantation in cardiac repair

چکیده مقاله:
Objective(s):Although previous studies have confirmed the beneficial effects of human umbilical cord matrix stem cell (hUCM) transplantation post myocardial infarction (MI), but this stem cell resource has no potential to induce angiogenesis. In order to achieve the process of angiogenesis and cardiomyocyte regeneration, two required factors for cardiac repair agents were examined namely; hUCM and VEGF on an infarcted heart. The main objective of this research is to investigate the combinatory effect of dhUCM and VEGF transplantation on an infarcted heart. Materials and Methods:45 min of ligating the left anterior descending coronary artery, the MI-induced animals received 50 μl PBS, 5 μg VEGF, 5×106 hUCM cells alone, combined with 5 μg VEGF and 5×106 differentiated hUCM cells alone or combined with 5 μg VEGF through intramyocardial injection. MI group, without hUCM and VEGF served as the control group. Left ventricular function and angiogenesis were also evaluated. Results: After eight weeks post MI, there were significant rise in left ventricular ejection farction in dhUCM+VEGF group compared to the other treated and non-treated groups (P

متن کاملHypoxia Preconditioning Promotes Survival And Clonogenic Capacity Of Human Umbilical Cord Blood Mesenchymal Stem Cells
Background: In recent decade, human umbilical cord blood derived mesenchymal stem cells (hUCB-MSCs) provide enormous potential for appropriate cell therapy, but they have limited growth potential and cease to proliferate due to cellular senescence, so providing a strategy for increasing the stem cell survival is necessary. Methods: In this investigation, MSCs characterized by flow cytome...
متن کاملP 82: The Transplantation of Human Umbilical Cord Mesenchymal Stem Cells in Neonatal Strokes
Brain injuries that caused by strokes (result of intra partum ischemia) are a frequent cause of prenatal mortality and morbidity with limited therapeutic options. Transplanting human mesenchymal stem cells (hmscs) indicates improvement in hypoxic Ischemic brain injury (HIBD) by secretion growth factor stimulating repair processes (Hmscs) known as multi potent cells which isolated from bone marr...
متن کاملIn Vivo Tracing of Human Umbilical Cord Matrix Stem Cells Useing MRI
Purpose: Human umbilical cord matrix (UCM) (Wharton jelly) stem cells labeling are tracking by MRI. Materials and Methods: After 48 hours incubation with USPIO human umbilical cord matrix (UCM) stem cells were labeled with USPIO by the means of receptor-mediated endocytosis. Prussian blue staining and Atomic absorption spectroscopy were performed to identify and show the iron oxide nanoparticle...
متن کاملHuman umbilical cord mesenchymal stem cell transplantation restores damaged ovaries
Ovarian injury because of chemotherapy can decrease the levels of sexual hormones and potentia generandi of patients, thereby greatly reducing quality of life. The goal of this study was to investigate which transplantation method for human umbilical cord mesenchymal stem cells (HUMSCs) can recover ovarian function that has been damaged by chemotherapy. A rat model of ovarian injury was establi...
